Abstract INTRODUCTION: In Brazil, West Nile virus (WNV) was first detected, in 2018, in horses with neurological disease. AIM: We report the first case of WNV infection in a horse from Ceará state and the complete genome sequence of an isolate from Espírito Santo state. Both infections occurred in 2019. METHODS: WNV was isolated from the tissues of a horse with neurological signs in Espírito Santo and sequenced by MiSeq. RESULTS: Phylogenetic analysis revealed that the isolate belongs to lineage 1a, clustering with the NY99 strain, a strain that has not circulated in the USA since 2005. CONCLUSIONS: Our findings reinforce the hypothesis that WNV has been silently circulating in Brazil for many years.

RESUMO: Pseudorraiva (PR) é uma doença viral altamente contagiosa de grande importância sanitária e econômica na indústria suína. O objetivo deste estudo foi avaliar diferentes regiões genômicas, químicas de PCR em tempo real e equipamentos para o diagnóstico molecular de PR. Foram avaliados quatro genes (GB, GC, GE, gD), três diferentes produtos químicos (SybrGreen, sondas de hidrólise e plexor) e dois equipamentos (ABI7500, Rotorgene 3000). Oligonucleotídeos com alvo para gB baseados na química de sondas de hidrólise apresentaram o melhor desempenho nos testes de eficiência (99%), de limite de detecção (10-1.5 TCID50 mL-1) e sensibilidade diagnóstica. Portanto, estes foram selecionados para fatores de verificação de desempenho, tais como a repetibilidade, reprodutibilidade e robustez (1,39% de variância entre dias, 24% variância entre analistas e 4,07% de variância por erro de análise). A qPCR, padronizada e validada neste trabalho, mostrou-se confiável para o diagnóstico de PR e pode ser utilizada em laboratórios de diagnóstico que se seguem normas internacionais como ISO 17025 e ISO 16140.

ABSTRACT: Pseudorabies (PR) is a highly contagious viral disease of great animal health and economic importance in swine industry. The aim of this study was to evaluate different genomic regions, real-time PCR chemistries and equipment for the molecular diagnosis of PR. Eight primer pairs targeting four genes (gB, gC, gE, gD), three different qPCR chemistries (SybrGreen, hydrolysis probes and plexor) and two equipment (ABI7500, Rotorgene 3000) were evaluated. Oligonucleotides targeting gB using hydrolysis probes showed the best performance after evaluating efficiency (99%), the detection limit (10-1.5 TCID50 mL-1) and diagnostic sensitivity and; therefore, those primers were selected for performance verification factors such as repeatability, reproducibility and robustness (1.39% variance between days, 24% variance between analysts and 4.07% variance in analysis error). The qPCR standardized and validated in this research proved to be reliable for the diagnosis of PR and may be used in diagnostic laboratories that follow ISO 17025 and ISO 16140.

O objetivo deste trabalho foi realizar a validação de uma reação em cadeia da polimerase em tempo real com o sistema Plexor® (qPCR) para o diagnóstico da Leucose Enzoótica Bovina (LEB), por meio da comparação com testes de diagnóstico recomendados pela Organização Mundial de Saúde Animal (OIE). A qPCR foi comparada com duas outras técnicas: a PCR nested (nPCR) e a imunodifusão em gel de ágar (IDGA). Das 82 amostras analisadas pela qPCR e nPCR, 79 apresentaram resultados concordantes, sendo a concordância, classificada pelo Índice Kappa, como alta. Entre as PCRs e a IDGA, o número de resultados concordantes foi de 71 e 69, respectivamente, para qPCR e nPCR, sendo a concordância classificada como considerável. A qPCR apresentou altos valores de sensibilidade e especificidade. Os valores preditivos da qPCR observados demonstraram a alta capacidade de classificação dos casos positivos e negativos. A qPCR não foi capaz de detectar três amostras positivas e tem custo ligeiramente superior que a nPCR. Entretanto, a qPCR é uma técnica mais rápida, menos susceptível a contaminações, tem alta sensibilidade, não utiliza e não gera resíduos carcinogênicos. Concluímos que a qPCR pode substituir a nPCR recomendada pela OIE no diagnóstico de rotina em áreas em que a LEB é endêmica, como no Brasil.

The goal of this research was to validate a Plexor® real time Polymerase Chain Reaction (qPCR) for Enzootic Bovine Leukosis (EBL) diagnosis by comparison with methods recommend by the World Animal Health Organization (OIE). The qPCR was compared with two other techniques: the nested PCR (nPCR) and to the agar gel immunodiffusion (AGID). Of 82 qPCR and nPCR analysed samples, 79 presented concordant results, being the concordance classified by Kappa Index as high. Between the PCRs and AGID, the number of concordant results was 71 and 69, out of 82, to qPCR and nPCR, respectively, being the concordance classified as considerable, in both cases. The qPCR presented high specificity and sensitivity values. The observed qPCR negative and positive predictive values show that the qPCR has a high capacity to correctly classify positive and negative results. The qPCR was not able to detect three positive animals and has a lightly higher cost than the nPCR. However, the qPCR its faster, less prone to contamination, has a high sensitivity and does not use or generate carcinogenic residues. We conclude that the qPCR may be used to replace the OIE nPCR technique in routine diagnosis in areas where EBL is endemic, as is the case of Brazil.

A reação em cadeia pela polimerase (PCR) foi utilizada para a detecção do vírus da leucemia bovina (VLB) em leucócitos periféricos de bovinos infectados. Os iniciadores utilizados foram construídos para amplificar uma parte do gene env do VLB. Os produtos da PCR foram analisados por eletroforese em gel de agarose corados por brometo de etídeo. A especificidade analítica da PCR foi confirmada por restrição enzimática dos produtos da reação com Bam HI e também pela análise da seqüência de três amostras. Sessenta e cinco animais foram testados para a presença de anticorpos anti-VLB, pela imunodifusão em gel de agar (IDGA) e pela PCR, para detecção direta do VLB. Houve 73,80% de concordância entre os dois testes. Quatro animais positivos na IDGA foram PCR negativos, enquanto 13 animais negativos na IDGA foram positivos na PCR. A sensibilidade diagnóstica obtida foi de 0,87 e a especificidade diagnóstica 0,62. A PCR desenvolvida pode ser uma ferramenta complementar no diagnóstico de infecções causadas pelo VLB, mas deve ter sua sensibilidade diagnóstica melhorada.

Polymerase chain reaction (PCR) was used for bovine leukemia virus (BLV) detection in the peripheral leukocytes of the infected bovines. The primers used were designed to amplify a part of env gene of BLV. PCR products were analyzed by agarose gel electrophoresis stained by ethidium bromide. The analytical specificity of PCR was confirmed by enzymatic restriction analysis of the PCR product with Bam HI and also by nucleotide sequence analysis of three PCR samples. Sixty five animals were tested for anti-BLV antibody, by agar gel-immunodiffusion test (AGID) and for direct BLV detection by PCR. There was a 73.80% concordance rate between the two tests. Four animals positive in AGID were PCR negative, while 13 AGID negative animals were found PCR positive. PCR got a 0.87 diagnosis sensitivity and 0.62 specificity. The developed PCR may be complementary tool in the diagnosis of BLV infection, but should have it diagnosis sensitivity improved.
